The peptide corresponding to amino acids 257-382 that contains the second bromodomain sequences of BRDT (accession number NP_997072.1) was expressed in E. coli and contains an N-terminal GST tag with an observed molecular weight of 42.8 kDa.
BRDT is similar to the RING3 protein family. It possesses two bromodomain motifs and a PEST sequence (a cluster of proline, glutamic acid, serine, and threonine residues), characteristic of proteins that undergo rapid intracellular degradation. The bromodomain functions as a 'reader' of epigenetic histone marks and regulates chromatin structure and gene expression by linking associated proteins to the recognized acetylated nucleosomal targets.
