ELISA: Indirect: To detect hsCD40-Ligand by indirect ELISA (using 100 μL/well antibodysolution) a concentration of 0.5 - 2.0 μg/mL is required. In conjunction with compatiblesecondary reagents, it allows the detection of at least 0.2 - 0.4 ng/well of recombinanthsCD40-Ligand. Sandwich: To detect hsCD40-Ligand by sandwich ELISA (using 100 μL/well antibodysolution) a concentration of 0.5 - 2.0 μg/mL is required. In conjunction with BiotinylatedAnti-Human sCD40-Ligand as a detection antibody, it allows the detection of at least0.2 - 0.4 ng/well of recombinant hsCD40-Ligand. Western Blot: To detect hsCD40-Ligand by Western Blot analysis this antibody can be usedat a concentration of 0.1 - 0.2 μg/mL. Used in conjunction with compatible secondaryreagents the detection limit for recombinant hsCD40-Ligand is 1.5 - 3.0 ng/lane, undereither reducing or non-reducing conditions.
Other applications not tested.
Optimal dilutions are dependent on conditions and should be determined by the user. -

背景
- CD40 ligand (CD40L) is a 33 kDa type II membrane glycoprotein expressed mainly on the cell surface of activated T lymphocytes, but also exists as a soluble form extracellularly. CD40L is the ligand for CD40, a member of the TNF superfamily, which is expressed on the cell surface of B cells, macrophages/monocytes, dendritic cells, vascular endothelial cells, and epithelial cells. CD40L plays an important role in B cell proliferation, antibody class switching, modulation of apoptosis in the germinal center through interaction with B cells expressing CD40, and activation of CD4+ T cells. Mutation within the CD40L gene is linked to hyper IgM syndrome, an X linked immunodeficiency disease that is characterized by elevated level of serum IgM and decreased level of other isotypes.Synonyms: CD40 ligand, CD40-L, CD40LG, GP39, T-cell antigen Gp39, TNF-related activation protein, TNFSF5, TRAP, Tumor necrosis factor ligand superfamily member 5
