- Armenian hamsters were immunized with recombinant mouse TIGIT tetramers by a combination of s.c. and food pad immunization and booster injections. Draining lymph nodes were fused with Sp2/0-Ag14 cells, selected in HAT (hypoxanthine/aminopterin/thymidine) medium and supernatants were screened for specificity by ELISA and flow cytometry using TIGIT-transfectants.

- 4D4 is an agonistic anti-TIGIT antibody, and addition of plate bound 4D4 to anti-CD3/anti-CD28 stimulated T cells was shown to inhibit their proliferation (Joller 2011). Further, 4D4 has been used in order to identify genes that are downstream of TIGIT signaling in Tregs, as TIGIT engagement might modulate the Treg phenotype in tumor tissue (Liu 2016).
