For immunohistology and Western blotting dilutions to be used depend on detection system applied. It is recommended that users test the reagent and determine their own optimal dilutions. The typical starting working dilution is 1:10. For selection of the most useful dilution in a given situation a test staining with cells or tissue known to express the antigen should be performed. To this end a small fresh skin biopsy can be incubated for 4 hours with TNF-alpha (1 ng/mL), IL-1 (100 U/mL) or LPS (1 μg/mL) in tissue culture medium at 37° C.
The antibody reacts with both intact human ICAM-1, CD54 and with soluble human ICAM-1. The antibody reacts also with ICAM-1 present on chimpanzee, rhesus monkey, cynomolgus monkey and baboon tissues.
