    背景

Protein Background: Protein kinase C (PKC) is a family of serine- and threonine-specific protein kinases that can be activated by calcium and the second messenger diacylglycerol. PKC family members phosphorylate a wide variety of protein targets and are known to be involved in diverse cellular signaling pathways. PKC family members also serve as major receptors for phorbol esters, a class of tumor promoters. Each member of the PKC family has a specific expression profile and is believed to play a distinct role in cells. PRKCD (Protein Kinase C Delta), a calcium-independent, phospholipid- and diacylglycerol (DAG)-dependent serine/threonine-protein kinase that plays contrasting roles in cell death and cell survival by functioning as a pro-apoptotic protein during DNA damage-induced apoptosis, but acting as an anti-apoptotic protein during cytokine receptor-initiated cell death. PRKCD is involved in tumor suppression as well as survival of several cancers and is required for oxygen radical production by NADPH oxidase and acts as positive or negative regulator in platelet functional responses. PRKCD negatively regulates B cell proliferation and also has an important function in self-antigen induced B cell tolerance induction. Upon DNA damage, PRKCD activates the promoter of the death-promoting transcription factor BCLAF1/Btf to trigger BCLAF1-mediated p53/TP53 gene transcription and apoptosis. Diseases associated with PRKCD include Autoimmune Lymphoproliferative Syndrome, Type Iii and Autoimmune Lymphoproliferative Syndrome.

Short Description: Recombinant PRKCD protein was expressed in baculovirus expression system as the full length protein (accession number NP_001303256.1) with a N-terminal FLAG tag. The molecular weight of the protein is 78.8 kDa.
