Competitive ELISA

用于多种样本类型中小分子、肽类和激素定量检测的竞争性ELISA。 在竞争性ELISA中，信号强度与抗原浓度呈反比关系：分析物浓度越高，检测信号越低。

该方法基于样本中抗原与包被在酶标板上的抗原之间对有限抗体的竞争结合。首先将未标记的一抗与样本共同孵育，然后将混合物加入预包被相同抗原的孔中。样本中抗原含量越高，可用于结合板上抗原的抗体就越少。

洗涤后加入与一抗特异结合的酶标二抗，并加入底物显色或产生荧光信号。最终信号随抗原浓度增加而降低。

当无法获得合适的抗体配对，或目标分子过小无法同时结合捕获抗体和检测抗体时，竞争性ELISA是理想选择。 因此该方法常用于小分子、肽类以及部分激素的检测。
