25 μg of whole-cell extracts from unstimulated 293T/17 cells and HeLa cells stimulated with 5 ng/ml EGF for 2 minutes per well were assayed using the Ras GTPase Chemi ELISA Kit (Figure 1). For lot 05715007, the signals were read on a luminometer within 15 minutes of adding the chemiluminescent reaction buffer. For the luminometer, the gain was set at 150 and the integration time was set to maximum. The level of Ras activation, and the ratio between the stimulated and unstimulated samples may vary depending on the cell type tested and the treatment used.
