The DNA is MseI digested and hybridized to the provided LINE-1 probe according to the product manual instructions for capture of the Long Interspersed Nucleotide Element 1 repeat elements of human genomic DNA. The provided methylated and non-methylated DNA standards are used to generate a standard curve for DNA quantification. The OD 450nm results of Lot 11115003 with a 2.5 minute developing time are shown in Figure 1. The data is then blanked and the net readings are graphed using a quadratic equation to generate a standard curve for quantification (Figure 2). Results show a reduction in global DNA methylation for the 5-azacytidine-treated HeLa cells as expected.
