TMEM108 (Transmembrane Protein 108) is a transmembrane protein originally identified as Retrolinkin, a protein involved in retrograde axon transport in the brain.1
TMEM108 is predominantly expressed in the brain where it has roles in neural connectivity and plasticity.
Indeed, TMEM108 has garnered attention in neuropsychiatric research due to its involvement in several cellular processes critical for proper brain function. A growing body of evidence links TMEM108 dysregulation to a range of neuropsychiatric disorders, including schizophrenia, bipolar disorder and major depressive disorder (MDD).2,3
TMEM108 plays a role in dendritic spine formation and synaptic transmission in neurons. Abnormalities in these processes can disrupt neural circuitry, which is a common feature in neuropsychiatric conditions. Disrupted synaptic connectivity has been implicated in the cognitive and behavioral symptoms observed in disorders such as schizophrenia.4,5
Beyond neurons, TMEM108 is expressed in oligodendrocytes and their precursors. Impaired oligodendrocyte function and myelin integrity can affect signal propagation along axons, potentially contributing to the white matter abnormalities frequently reported in psychiatric disorders. This dual role in both neuronal and glial cells positions TMEM108 as a key factor in maintaining overall brain network integrity.6
Given its multifaceted role in brain physiology, TMEM108 is being investigated as a potential biomarker for early diagnosis and prognosis in neuropsychiatric disorders. Understanding its function could also pave the way for developing targeted therapies aimed at restoring synaptic and myelin integrity.
