ATG9A is present in numerous intracellular compartments, including the trans-Golgi network and early and recycling endosomes, and traffics through the secretory pathway to the plasma membrane and to the endocytic pathway from the plasma membrane. ATG9A protects cells against plasma membrane damage caused by a spectrum of exogenous and endogenous agents, including permeabilization by gasdermin and mixed lineage kinase domain like (MLKL), which generate pores at the plasma membrane or perturb plasma membrane integrity, respectively, during programmed cell death processes of pyroptosis and necroptosis. We furthermore define the ATG9A-IQGAP1 apparatus that integrates with the ESCRT system to cooperatively heal areas of plasma membrane damage.
