Description: Neuronal pentraxin 2 (NP2), also known as NPTX2, NP-II, and neuronal activity-regulated pentraxin (Narp), is a calcium-dependent lectin secreted protein member of the pentraxin family involved in synaptic plasticity and excitatory homeostasis.1,2NP2 is part of the neuronal pentraxin family that includes two other members: Neuronal Pentraxin 1 (NP1) and Neuronal Pentraxin Receptor (NPTXR). The pentraxin superfamily members are arranged in pentameric structures with a discoid shape. The family members are characterized by a 205 amino acids long conserved sequence located at C-terminal called the pentraxin domain 2,3.NP2 is widely expressed in the brain, especially in the hippocampus, dentate gyrus, cerebellum, and cerebral cortex regions, as well as in the testis, pancreas, liver, heart, and skeletal muscle.2,3NP2 interacts with α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptors, which are crucial for excitatory neurotransmission in the brain. By binding to AMPA receptors, NP2 influences their trafficking and localization at synapses, affecting the strength of excitatory synaptic transmission 2,3.Given its presence in brain regions associated with learning and memory, alterations in NP2 levels or function have been implicated in cognitive processes. Dysregulation of NP2 has been observed in various neurological disorders, suggesting its potential importance in maintaining cognitive function.4-6Abnormalities in NP2 expression have been linked to neurodegenerative disorders, including Alzheimer's disease. Changes in NP2 levels may contribute to synaptic dysfunction and cognitive decline observed in these conditions.4-6
