Approved: ELISA (0.5 - 1 μg/mL), ICC (10 μg/mL), IHC (10 μg/mL), WB (0.1 - 0.2 μg/mL)
Usage: Suitable for use in ELISA, Western Blot, Immunocytochemistry. Western Blot: 0.1-0.2 μg/mL with the appropriate secondary reagents to detect mouse rmLipin 3. The detection limit for rmLipin 3 is approximately 2 ng/lane under non-reducing and reducing conditions. Immunocytochemistry: 10 μg/mL to detect Lipin 3 on CCD-18co normal human colon fibroblasts. Cells were fixed with PBS containing 4 % paraformaldehyde for 20 minutes at RT and blocked with PBS containing 10 % normal donkey serum, 0.1 % Triton X-100, and 1 % BSA for 45 minutes at RT. After blocking, cells were incubated with diluted primary antibody overnight at 4?C followed by donkey anti-goat IgG at RT in the dark for one hour. Between each step, cells were washed with PBS containing 0.1 % BSA. Immunohistochemistry: 10 μg/mL to detect Lipin 3 in cryosections of adult mouse intestine. Tissue was fixed overnight in 4 % PFA + 14 % picric acid in PBS at 4?C. Sections were blocked with 10 % donkey serum and 0.1 % Triton in 1 % BSA in PBS for 45 minutes at RT. After blocking, sections were incubated with primary antibody for 3 hours at RT followed by donkey anti-goat lgG at RT for 1 hour. Between each step, sections were washed with 0.1 % BSA in PBS. Direct ELISA: 0.5-1.0 μg/mL with the appropriate secondary reagents to detect mouse rmLipin 3. The detection limit for rmLipin 3 is approximately 0.05 ng/well. -
