Approved: ELISA (0.5 - 1 μg/mL), ICC (10 μg/mL), IHC (10 μg/mL), WB (0.1 - 0.2 μg/mL)
Usage: Suitable for use in ELISA, Western Blot, Immunocytochemistry and Immunohistochemistry. Western Blot: 0.1-0.2 μg/mL. Detection limit for rhVersican is ~2 ng/lane under non-reducing and reducing conditions. Immunocytochemistry: 10 μg/mL detects Versican in Hela cells. Cells were fixed with PBS containing 4 % paraformaldehyde for 20 minutes at RT and blocked with PBS containing 10 % normal donkey serum, 0.1 % Triton X-100, and 1 % BSA for 45 minutes at RT. After blocking, cells were incubated with diluted primary antibody overnight at 4? C followed by fluorochrome-coupled anti-goat IgG at RT in the dark for one hour. Between each step, cells were washed with PBS containing 0.1 % BSA. Immunohistochemistry: 10 μg/mL detects Versican in cryosections of E13.5 mouse embryos. Tissue was fixed with PBS containing 4 %PFA for 20 min. at RT and blocked with PBS containing 10 % normal donkey serum, 0.1 % Triton and 1 % BSA for 45 minutes at RT. After blocking, sections were incubated with diluted primary antibody for 3 hours at RT followed by fluorochrome-coupled anti-goat lgG at RT for 1 hour. Between each step, sections were washed with 0.1 % BSA in PBS. Direct ELISA: 0.5-1.0 μg/mL. Detection limit for rhVersican is ~0.1 ng/well. Optimal dilutions should be determined by each laboratory. -
