- RCE1 selective antibodies are generated against a peptide taken from the C-terminal region spanning AA 310-329. The RCE1-selective antibodies are affinity purified on an immobilized antigen based affinity matrix, the isolated antibodies are then stabilized in antibody stabilization buffer for long-term storage. The AnxA10-selective antibodies are fully characterized for applications in western blotting and ELISA at the recommended dilutions. The Supplier provides RCE1 Western blot positive control samples in SDS-PAGE sample buffer.

背景
- Oncogenic protein RAS-converting enzyme 1 (RCE1) is an endoprotease which is involved in prenylation or posttranslational modification of a carboxyl-terminal cysteine residue of proteins that terminate with a CAAX motif such as the nuclear lamins and the RAS family of proteins. After prenylation, the last three residues of CAAX proteins are clipped off by an endoprotease (like RCE1) of the endoplasmic reticulum and the prenylcysteine residue is methylated. Prenylation is essential for the proper intracellular targeting and function of several CAAX motif containing proteins. RCE1 gene encodes an integral membrane protein which is a member of the metalloproteinase family. Studies indicate that the endoproteolytic processing of CAAX proteins is essential for cardiac functions. The heart-specific RCE1 knockout mice but not liver specific-RCE1 knockouts have dilated cardio myopathy with normal liver function. Inactivation of RCE1 mouse fibroblasts inhibits RAS transformation by mislocalization of RAS proteins away from plasma membranes. The inactivation of RCE1 also exacerbates myeloproliferative disease by increasing the flow of immature hematopoietic cells in to circulation and infiltration of cells into liver and spleen leading to death. RCE1 is an important component of the RAS activation pathway and is a potential target for therapeutic intervention. RCE1 gene is 329 AA (40 kDa) and is localized on human chromosome 3q11.
