产品特性 Tris Buffer, after dilution, is used on formalin-fixed, paraffin-embedded tissue sections mounted on glass slides for target retrieval prior to immunohistochemistry (IHC) procedures. When performed using this reagent and elevated temperature, target retrieval can disrupt the covalent bonds formed by formalin in tissue. Removal of these bonds can allow renaturation of protein molecules and increased antibody accessibility, which in turn can result in increased antibody binding (and thus increased staining intensity) and improved signal to noise ratios. This type of result is observed with many primary antibodies.
实验流程 1. Deparaffinize and rehydrate tissue sections.
2. Dilute the product 1:100 with deionized water. Example 2 ml of concentrate diluted in 198 ml of deionized water.
3. Immerse slides in a coplin jar filled with the diluted Tris Buffer reagent. Microwave on HIGH power until boiling.
4. Keep the slides warm by heating for 10 minutes on LOW power.
5. Let the coplin jar sit in microwave for at least 20 minutes.
6. Remove and rinse with water.
7. Rinse with buffer and proceed with the appropriate staining protocol for the primary antibody in use.
